Iodomethane-d3 is a colorless liquid, which can be used as isotope labeling reagent. It is a useful research chemical, which is used in biochemical research to analyze peptides and glycans by analytical spectroscopy. Deuterated amino acids have the same structure and similar properties with common amino acids. Deuterated amino acids can be widely used as tracers and internal standards. IODOMETHANE-D3 (methyldeuterium iodide) has a wide range of applications in the laboratory.

1. Labeling reagent: It can be used as a labeling reagent to label hydrogen atoms in compounds. Due to its presence of deuterium isotopes, compounds labeled with it can be more easily detected and identified through mass spectrometry and other techniques compared to naturally abundant hydrogen isotopes (with a relative abundance of 0.0156% deuterium isotopes).

2. Reference standard substance for nuclear magnetic resonance (NMR): Due to the clear signal of product in the NMR spectrum, it is commonly used as a reference standard substance for deuterium spectroscopy (2H NMR). The characteristic chemical shift signal can be used to calibrate the sample solvent and analyze and identify the NMR spectra of other compounds.

3. Reaction reagent: it can be used as a reaction reagent in synthetic chemistry. It can react with alkali metals, boron alcohols, haloalkanes, etc. to generate compounds with deuterium labels. These labeled compounds can be used to study reaction mechanisms, track reaction pathways, and analyze and characterize the properties of compounds.

4. Solvent: it can be used as an inert solvent, especially suitable for certain high sensitivity experiments. Most deuterated iodomethane is prepared through the iodization reaction of deuterated methanol, and different synthesis methods can be used depending on the type of iodization reagent.

Method 1:

Slowly inject red phosphorus reagent (50.0 g, 1.6 mol), H2O (100 mL), and elemental iodine (250.0 g, 1.0 mol) into a dry 250 mL flask with a reflux condenser at -15 ° C for 0.5 hours. Then gradually add deuterated methanol (30.0 grams, 0.8 moles) to the reaction mixture. Heat the reaction mixture to 65 ℃ and continue stirring for about 2 hours. After the reaction, cool the reaction mixture to room temperature. Finally, distill the reaction mixture at 45 ℃ and collect the corresponding fractions to obtain the target product molecule D3 iodomethane.

Method 2:

Add deuterated methanol (CD3OD) (0.5 g, 1.13 mL, 0.0138 mol) to dry dichloromethane (10 mL) in a 20 mL round bottom flask. Then add TMSI (2.77 grams, 1.98 milliliters, 0.0138 moles) to the obtained reaction solution, and slowly stir the reaction mixture at 0 ℃ for several hours. Then transfer it to room temperature and stir the reaction mixture at room temperature for 8 hours. After the reaction is completed, there is no need for further purification. The deuterated iodomethane obtained from the reaction can be directly used for the next step of the reaction.
